Synopsis "L-Lactid-Copolymere für die Insulinverabreichung (in German)"

Neuartige Triblock-Copolymere aus Poly(L-Lactid)-Poly(Ethylenglykol)-Sebacat-Poly(Ethylenglykol)-Poly(L-Lactid) wurden durch Ringöffnungspolymerisation (ROP) synthetisiert. Zwei Gruppen von achtzehn Polymernetzwerken wurden ebenfalls hergestellt; die ersten neun Netzwerkgruppen wurden aus Triblock-Copolymeren mit vernetztem Natriumalginat hergestellt, S1-S9. Zur Herstellung der zweiten Gruppe von neun Netzwerken, SP1-SP9, wurde Pluronic F-127 zu den vorherigen Zusammensetzungen hinzugefügt. Alle diese Polymernetzwerke wurden mit 50 IU Insulin beladen. Es wurde eine Studie zur Insulinfreisetzung bei einem pH-Wert von 1,2 und 8,4 über 24 Stunden durchgeführt, die ergab, dass die Freisetzung aus kinetischer Sicht der ersten Ordnung folgt. Die erhaltenen Daten zeigten, dass die Insulinfreisetzung innerhalb der ersten zwei Stunden bei Faser-IPNs mit einem Molekulargewicht von 2000 g/mol Poly(ethylenglykol) und 50 Einheiten Poly(L-Lactid) abnimmt.
