Synopsis "Synthesis of tyrosinase by streptomyces SP."
- In all 15 isolates of Streptomyces were screened for the synthesis of tyrosinase by both qualitative and quantitative methods in synthetic medium. - Activity of tyrosinase, specific activity of tyrosinase, formation of melanin and protein content of tyrosinase were assessed to determine the catalytic power of tyrosinase. - In a batch wise bioprocess, an isolate of Streptomyces DLS-8 has been found to be potential for the production of tyrosinase. - Effect of pH and temperature on the production of tyrosinase were also examined. 300C temperature and pH 8.5 were proved to be optimum for the maximum production of tyrosinase. - Results of the present study revealed that starch and citrulline were the important carbon and nitrogen sources respectively for the maximum production of tyrosinase. - The partial isolation and purification of the tyrosinase was under taken by SDS-PAGE. The tyrosinase was determined to be about 45 KDa, comparing to BSA as standard marker.- Liquid chromatogram mass spectrum reveals the presence of tyrosine, L-Dopa and melanin based on their base peaks for their respective molecular weights indicating significantly the catalytic power of tyrosinase.
