Synopsis "DNA Barcoding: Philippinische Cinnamomum Schaeffer (Lauraceae) (in German)"

Die 27 gesammelten Arten wurden einer DNA-Isolierung, einer Polymerase-Kettenreaktion und einer DNA-Sequenzierung unterzogen. Eine modifizierte CTAB isolierte erfolgreich die DNA der Arten. Die Amplifikation mit rbcL und Master-Mix lieferte 36 gute DNA aus 45 Proben. Die BLAST-Ergebnisse ergaben eine maximale Identität von 86 % bis 100 %. Die MEGA 7-Analyse führte zu Cluster 1, der aus 34 Individuen in 22 Arten bestand, mit 75 % Bootstrap-Unterstützung. Cluster 2 besteht aus zwei Arten als zwei Singletons.Bootstrap-Zweig sind alle über 50%, niedrigsten 60% und höchsten von 75 %.Einige der Arten zeigten genetische Divergenz.Die 36 DNA-Proben gehören zu nur 14 Haplotypen in 140 Nukleotiden sites.Haplotype 1 hat 18 Proben von 12 Arten.It bestätigt, dass alle Individuen sind Cinnamomum Arten. Ähnliche Arten, die an verschiedenen geografischen Standorten vorkommen, weisen leichte Unterschiede in den DNA-Sequenzen auf.
